愈心痛对犬缺血心肌细胞线粒体超微结构损伤保护作用的体视学研究  

Study on the Protective Effects of YXT on Mitochondrial Ultrastructural Damage of Ischemic Myocardium by Cell Morphometry

摘　　要：本实验在结扎麻醉犬冠状动脉左前降支中段造成急性心肌缺血的模型上，动态检测了犬血清肌酸磷酸激酶（ＣＰＫ）活性变化，并运用电镜技术产体视学研究方法对缺血心肌细胞线粒体的超微结构损伤进行了形态定量分析，以探讨具有益气活血功效的中药复方愈心痛胶囊对急性心肌缺血的保护作用。心肌酶检测显示：心肌缺血时间越长，血清ＣＰＫ活性越高，而愈心痛可阻止缺血心肌ＣＰＫ的漏出，使其活性显著低于生理盐水对照组；电镜体视学研究表明：缺血心肌细胞线粒体的体密度（Ｖｖ）明显增加，面数密度（ＮＡ）和比表面（δ）却降低，提示线粒体高度水肿，而给予愈心痛后可显著降低缺血心肌线粒体的Ｖｖ，同时增加ＮＡ和δ，表明愈心痛胶囊可减轻缺血造成的心肌线粒体水肿，阻止缺血心肌膜损伤，有稳膜作用，在一定程度上维持膜结构的完整性，从而保护心肌超微结构，防治心肌缺血性损伤。提示愈心痛胶囊对缺血性心脏病的治疗具有潜在的临床应用价值。In this experiment,the protective effects of Yuxintong(YXT) on myocardial ischemic injury were studied.The acute myocardial ischemia models were reproduced in anesthetized dogs by ligating the left coronary artery at the middle of anterior descending branch.The serum creatine phosphokinsse (CPK) activity was monitored after coronary occlution in different times,and then the ultrastructural degenerative changes of the dog's myocardium were observed after 180 minutes ischemia and assayed by cell morphometry.Results showed that YXT capsule were able to effectively inhibit the leakage of CPK,alleviate the myocardial ultrastructural degenerative changes,reduce mitochondrial volume density and increase its special surface as well as numerical density on area,therefore,attenuating the damage of mitochondrial ultrastructure of dog's myocardium caused by acute ischemia.It suggested that YXT capsule had notable protective action on integrity of myocardial structure and cytomembrance.
