摘  要:【目的】研究缺血再灌注心肌的相关蛋白变化。【方法】将8只新西兰大白兔随机分为2组(n=4):正常心肌对照组(C组)和缺血再灌注心肌组(R组)。C组正常灌注160 min,不阻断左冠状动脉;R组左冠状动脉前降支阻断40 min,开放再灌注120 min。分别取各组左心室心肌进行二维凝胶电泳,利用ImageMaster 2D软件分析实验结果。【结果】R组和C组对比,有17个蛋白表达发生了显著变化,其中表达增强的有7个蛋白,表达降低的有4个蛋白,表达不匹配(仅在R组出现)的有6个蛋白点。【结论】这些差异表达的蛋白可能在心肌缺血再灌注损伤的保护中发挥作用。[Objective]To analyze the expression of proteome in myocardial tissue of ischemic-reperfusion rabbit. [Methods] Eight New Zealand white rabbits of both sexes were randomly assigned to group C and group R. Group R underwent ligation of the left anterior descending coronary artery (LAD) for 40 minutes and reperfusion for 120 minutes. Group C kept flowing for 160 minutes without ischemic-reperfusion. Two dimensional polyacrylamide gel electrophoresis(2-DE) was followed by computer assisted image analysis and used to screen changes in protein patterns of extract of myocardium from Group C and Group R. [Results] Compared with Group C, there were 17 protein spots displayed quantitative changes in expression in Group R(P〈0, 05), of which, 7 spots showed high expression , 4 spots decreased in abundance and 6 only expressed in group R. [Conclusion] These differential proteins may play an important role in protection of myocardial ischemic-reperfusion injury.
