摘  要:目的探讨冈田酸(OA)诱导NG108-15细胞tau蛋白过度磷酸化过程中PTEN蛋白水平的变化。方法20 nmol.L-1OA孵育NG108-15细胞不同时间(0,3,6,12,18,24,36,48 h),倒置显微镜观察细胞形态的变化,免疫印迹法观测各时间点Ser396位点磷酸化tau蛋白及PTEN蛋白水平的变化。结果NG108-15细胞随OA孵育时间的延长逐渐出现细胞胞体变圆,突起回缩,悬浮生长。细胞Ser396位点磷酸化tau蛋白水平于OA作用6 h后显著升高,18 h达高峰,之后逐渐下降,但48 h仍高于基础水平;PTEN蛋白水平于3 h开始上升,18h达高峰,之后逐渐下降,12、18、24 h 3个时间点PTEN蛋白水平显著高于基础水平,36、48 h时间点PTEN蛋白水平与基础值比较差异无显著性。结论OA诱导NG108-15细胞AD样发生中,PTEN蛋白水平可能升高。OBJECTIVE To study the change of PTEN protein in the okadaic acid(OA) induced of tau hyperphosphorylation in NG108 - 15 cells. METHODS NG108 - 15 cells were treated with 20 nmol· L^-1 OA for 0, 3, 6, 12, 18, 24, 36, 48 h, and then observed by inverted microscope. Western Blotting was used to detect the levels of phosphorylated tau at site Ser^396 and the change of PTEN in NG108 - 15 cells. RESULTS Ceils changed gradually after treated with 20 nmol · L^-1 OA and appeared typical morphology of Alzheimer’s disease(AD). Level of phosphorylated tau at site Ser^396 was increased after exposure of 6 h, reached the maximum level after 18h, and then gradually decreased, but still maintain higher than normal level at 48 h. Level of PTEN was increased after exposure of OA 3 h, arrived at top after 18 h, and then gradually declined; level of PTEN at 12, 18, 24 h was significandy higher than normal level, while there's no significance of difference between level of PTEN at 36, 48 h and base level. CONCLUSION Level of PTEN protein seems probably increased in OA induced AD -like NG108 - 15 ceils.
