摘  要:目的探讨芥子碱对新生血管生成的影响。方法将聚乙烯醇海绵盘随机分为生理盐水对照组和不同剂量芥子碱组,植入小鼠皮下,构建盘状血管生成模型,观察术后12d的血管生成情况及组织生成情况,以蛋白定量检测试剂盒检测在海绵组织中的可溶性蛋白含量,对海绵冰冻切片行苏木精-伊红(HE)染色,应用计算机图像处理分析系统分析海绵内组织生长情况。结果与生理盐水对照组比较,芥子碱大、中剂量组(10.0 mg.mL-1和7.5 mg.mL-1)海绵盘的湿重、可溶性蛋白含量和组织生长面积都明显减少(P<0.05)。结论芥子碱具有一定抑制血管生成的作用,且呈剂量依赖性。OBJECTIVE To study the influence of Sinapine on the formation of microvessels. METHODS Sterile polyvinyl alcohol sponge discs which were labeled as physiological saline group and sinapine groups were implanted into the subcutaneous part of rat. 12 days after injection, the tissues ingrowth in the implants were observed and the quantities of the soluble proteins in the sponge were detected by Bradford Protein Assay. The pathological section of sponge was stained with hematoxylin and eosin(HE). The tissue ingrowth in the implants was analysed by image analysis system. RESULTS Compared with physiological saline group, the weight of sponge discs, the quantifies of the soluble proteins and the areas of the tissues that ingrowth in the implants in Sinapine groups(7.5mg· mL^-1 and 10.0mg · mL^-1) had heavy inhibition of angiogenesis (P 〈 0. 05). CONCLUSION Sinapine can prevent the formation of microvessels and the effects depend on the concentration.
