摘  要:对分离自不同地域的粘帚霉属不同种类的生防菌株rRNA基因转录间区进行了克隆测序,用Clustal X软件进行自动排序,用Njplot程序构建系统进化树.供试26个粘帚霉菌株分在4个组,与传统形态分类结果一致,同一种类的不同来源的菌株差异不明显.研究表明,可以根据ITS区DNA序列差异对粘帚霉属进行种级分类,但不能用于区分种内不同地域的菌株之间的差别.Classification of Gliocladium spp. was mostly done based on the methods of morphology and presented some difficulties. Molecular technique was used, and 26 Gliocladium isolates were investigated by 5.8S rDNA amplification with ITS1 and ITS4 as primers, sequencing and blast analysis. Results showed that the isolates tested were divided into 4 groups, the species of different groups were closely homologous and were different from control fungi. The phylogenetic tree also revealed that the same species isolates from different regions were not differ obviously and would be included in the same group. ITS sequence analysis result was identical to that of morphological classification of these fungi. It was concluded that the method of rDNA ITS sequence analysis was a powerful tool for the classification of Gliocladium at species level.
