Identifiler体系在亲子鉴定中的应用分析

摘　　要：目的运用AmpFlSTR Identifiler PCR扩增试剂盒用于亲子鉴定126例,探索此试剂盒在亲子鉴定案例中的应用价值。方法采用荧光标记STR-PCR复合扩增、毛细管电泳基因分型技术,应用ABI 310遗传分析仪对126例亲子鉴定的个体进行15个STR位点的基因分型。结果在126例亲子鉴定中,否定亲权的25个案例中均至少有3个位点不符;认定亲权的101例中,亲子关系指数(RCP)均>99.98%,在认定的亲子关系中,有3例出现1个位点的基因突变。结论应用Identifiler体系的15个STR位点的基因鉴定,能成功地开展亲子鉴定,并显著提高了亲子关系指数。Objective 126 cases parentage testing was performed by using Identifiler Amplification Kit and to study their application value in parentage testing. Methods The technique of fluorescent dye labeling multiplex STR-PCR, capillary electrophoresis and DNA sequencer GeneScan, Genotyping was adopted in genotyping a sample of individuals in 126 cases parentage testing. Results Among the 126 cases of parentage testing, 25 cases of exclusive paternity were incoordinate with more than 3 loci. Among the 101 cases of determined paternal relation,the relative chance of Paternity（RCP） were greater than 99.98%. 1 mutant gene observed in 3 paternity testing cases. Conclusion The results suggest that Identifiler Kit can be applied to parentage testing, and it will increase the RCP obviously.
