摘 要:目的对鬼针草抗小鼠急性肝损伤的有效部位进行筛选,并初步探讨其对肝脏的保护作用。方法采用四氯化碳(CC l4)小鼠急性肝损伤模型,测定肝脏指数、肝功能酶、肝匀浆丙二醛(MDA)含量、过氧化物歧化酶(SOD)活性,并光镜下观察肝脏病理变化。结果与正常组相比,CCl4模型对照组肝脏指数、血清ALT、AST活性和肝组织MDA含量显著升高(P<0.01),肝脏SOD活性显著降低(P<0.01),肝脏出现不同程度的变性及坏死,肝小叶及肝细胞消失,肝脏病理学分级与正常组有显著差异(P<0.01)。鬼针草黄酮类组分可使肝损伤小鼠的肝脏指数、ALT、AST和MDA显著降低,并升高SOD活性。病理检查显示鬼针草能明显减轻CC l4引起的肝脏结构损害,其中尤以黄酮类组分疗效最佳(P<0.01)。结论鬼针草黄酮类组分有显著的抗肝损伤作用,可能是鬼针草保肝作用的主要有效部位。Objective To select the active principle extracted from Bidens bipinnata L that can protect aganist acute liver injury, and to explore the hepatoprotective effect. Methods Seventy male Kunming mice were randomly divided into the normal control group, the CC14 injury model group, the A composition of Bidens bipinnata L group (360 mg/kg) ,the B composition of Bidens bipinnata L group ( 180 mg/kg) ,the C composition of Bidens bipinnata L group ( 100 mg/kg,and the Bifendate ( 100 mg/kg) group. All the mice except for those in the normal control group had acute liver injury induced by carbon tetrachloride ( CC14 ) 10 ml/kg injected intraperitoneally. The animals were killed 18 h after CC14 administration. The liver index, activities of serum aspartate transaminase (AST) , alanine transaminase (ALT),the levels of malondialdehyde (MDA), superoxide dismutase (SOD) in liver homogenate, as well as the pathological changes of the liver were determined in all groups. Results In comparison to the normal con- trol group, the liver index, the level of serum ALT, AST and the content of hepatic MDA were increased significantly in CC14 injury model group (P 〈 0. 01 ), while the activities of hepatic SOD were significantly decreased ( P 〈 0. 01 ). And the liver histology showed necrosis and degeneration, the disappearance of hepatic lobules and hepatocyte, and the degree of liver pathology lesions increased significantly compared to the normal group( P 〈0. 01 ). The liver index and serum ALT in groups treated by Bidens bipinnatal were significantly lower than those in model group( P 〈 0. 01 ). The liver necrosis and degeneration induced by CC14 were also improved significantly by Bidens bipinnatal. Conclusion The flavonoids of Bidens bipinnata L have protective effects aganist acute liver injury induced by CC14 in mice ,which maybe the major active principle of hepatoprotective effect of Bidens bipinnata L.
