摘  要:目的 探讨槲皮素对先天性多药耐药胆囊癌细胞株GBC-SD多药耐药性(MDR)的逆转情况。方法 分别以10个浓度阿霉素(3.33~33.28μg/ml)及其与不同浓度槲皮素(0.1、0.05、0.025μmol/L)混合液培养GBC-SD,观察细胞生存率及半数抑癌浓度值(IC50)。结果 单用阿霉素培养时IC50为16.65μg/ml,细胞生存率分别为80.58%~26.18%。加用槲皮素培养后IC50分别为6.66、9.99和13.32μg/ml,两两比较,P均〈0.05,且均显著低于单用阿霉素者(P均〈0.05);细胞生存率均显著低于单用阿霉素者,且逆转能力呈浓度依赖性。结论 槲皮素可逆转GBC-SD的MDR。[ Objective ] To research whether the drug resistance of human gallbladder carcinoma GBC-SD cells can be reversed by quercetin and the reversal degree. [ Methods ] GBC-SD cells were cultured in different concentration of adriamycin(3.33- 33.28 μg/ml) and quercetin(0.1,0.05, 0.025 μmol/L) medium. Then observed the cells survival rate and ICs0. [ Results] IC50 and cells survival rate was 16.65 μg/ml and 80. 58% - 26.18% when gallbladder carcinoma GBC-SD cells were cultured with adriamycin only. However when different concentration of quercetin was added, IC50 was 6.66 μg/ml( 0.1 μmol/L), 9.99 μg/ml ( 0. 05 μmol/L) and 13.32 μg/ml ( 0. 025 μmol/L) ; the differences among 3 groups had statistical significance ( P 〈 0.05 ). The survival rates in different concentration quercetin groups were all lower than those in adriamycin only groups. And under the same concentration of adriamycin, the degree of reverse increased with the concentration of quercetin. [ Conclusion ] Quercetin has the ability to reverse the drug resistance of gallbladder carcino- ma cells, and the reversal has a positive relation with the concentration of quercetin.
