摘 要:目的探讨体外反搏对减轻高脂饲养猪产生动脉粥样硬化的效果及机制。方法选雄性乳猪16头,用高脂喂养建立动脉粥样硬化模型,对模型猪进行36h体外反搏治疗。对腹主动脉作苏丹Ⅲ染色,分析脂质浸润阳性率;用基因芯片分析猪的动脉血管内皮细胞基因的表达。结果腹主动脉苏丹Ⅲ染色图像定量分析阳性率,高脂组为6.49%±0.84%,高脂加反搏组为2.29%±0.46%,两组差异有统计学意义(P<0.05)。发现改变的基因:芯片1(反搏组Cy3/高脂组Cy5):整合素β10.06、CTGF0.28;芯片2(反搏组Cy3/高脂组Cy5):整合素β10.62、CTGF0.39。结论体外反搏可以下调内皮细胞整合素β1、CTGF基因的表达,降低血管內皮细胞对脂质的摄取,起到抗动脉粥样硬化的作用。Objective To examine the mechanisms of EECP treatment in reducing atherosclerotic vascular disease. Method Endothelial cell injury was induced in 16 male pigs by feeding with 4% cholesterol-enriched diet for 12 weeks. 16 Pigs were then randomly divided into two groups: EECP group (8 pigs) and the control group (8 pigs). EECP treatment was performed for 36h(2h-treatment once-a-day for 18 days). All pigs were provided with highcholesterol-fed until the end of EECP treatment. The abdominal aorta was stained by Sudan Ⅲ dye. Endothelial cells were harvested from the thoracic aorta. Endothelial cells from 4 EECP pigs were pooled, total RNA from 4 EECP pigs was labeled with Cy3, and the RNA from the 4 control pigs was labeled with Cy5. Cy3 and Cy5 labelled probes were hybridized in one cDNA microarray chip. Two chips were used for hybridization. The gene expression level of aortic endothelial cells was analyzed by cDNA microarray. Result The levels of plasma cholesterol were increased in pigs feeding with high-cholesterol-fed (P〈0.05). There was no significant difference in plasma cholesterol level in EECP group compared to the control group (P〉0.05). The ratios of Sudan Ⅲ stained area in celiac arota were 2.29%± 0.46% and 6.49%±0.84% in EECP group and hypercholesterolemia group, respectively (P〈 0.05). The expression of integrin-β1 was down-regulated in EECP (0.06 fold) compared with the control group (0.62 fold). The expression of connective tissue growth factor (CTGF) was down-regulated (0.28-fold, 0.39-fold) in EECP group (0.28 fold) when compared with the control group (0.39). Conclusion EECP may reduce endothelial injury in high-cholesterol-fed pigs. The effect is associated with the down-regulation of the gene expression of integrin-β1 and CTGF. High level of expression of CTGF in endothelial cells may be related to atherosclerosis. The results suggested that EECP may reduce cholesterol uptake and inflammatory in endothelial.
