摘  要:目的:探讨慢性心肌缺血血瘀证模型小型猪血中血栓素B2、6-酮-前列腺素F1α、TNF-α浓度与心功能等指标的变化及其相关性。方法:采用病证结合的方法即动物左冠状动脉前降支第一间隔支起始部远端放置Ameroid缩窄环的方法复制慢性心肌缺血血瘀证模型。小型猪随机分为假手术组、模型组。应用心脏超声心动图测定心功能和心室重构;应用放免法测定血中血栓素B2(ThromboxaneB2,TXB2)、6-酮-前列腺素F1α(6-keto-PGF1α,6-K-PGF1α)和TNF-α的浓度。结果:小型猪慢性心肌缺血后,血中TXB2、6-K-PGF1α、TNF-α浓度与假手术组相比都发生了明显变化。结论:TXB2与EDV呈负相关、与TNF-α呈正相关,TNF-α表达水平与EDV、ESV均呈负相关,随心功能的恶化,其表达量增高。说明血小板的激活程度与心功能有关,心功能越差,血小板的活性越高,血栓形成的危险性就越大,尤其易造成微血栓。Objective To inquire into the change of the concentration of TXB2, 6-Keto-PGF1α, TNF-α in blood plasma of miniature pig which was the syndrome of blood stasis model made by chronic myocardial ischemia and the relation between them. Methods We adopted the syndrome of blood stasis model of chronic myocardial ischemia by laying the Ameroid condensation ring surrounded the originate region of the first septum of animals' front-fall branch of left coronary artery as the integrate illness with syndrome. And the pigs were divided into uncontrolled group and model group randomly. We tested the cardiac function and venlricular cordis re-establishment by ultrasonic cardiogram, and tested the value of TXB2, 6-Keto-PGF1α. and TNF-α by radioimmunoassay in blood plasma. Results The concentration of TXB2, 6-Keto-PGF1α, TNF-α all obviouslychanged compared with model group made by chronic myocardial ischemia of miniature pig. Conclusion The value of TXB2 and EDV presented negative correlation, it and that of TNF-α presented positive correlation. The express of TNF-α and EDV, ESV presented negative correlation, and with the deteriorate of cardiac function, it increased. These showed that the level of activation of Pit were related with cardiac function. The worse of cardiac function, the higher of the activity of Pit, and the more of dangerous of the thrombosis, and easier of mini-thrombosis especially.
