摘  要:目的观察背根神经节P2X_3受体在大鼠坐骨神经慢性压迫性损伤(CCI)后表达的变化,探讨慢性神经痛的发生机制。方法24只雄性SD大鼠随机分成假手术组(S组)、坐骨神经结扎组(CCI组),各组分别均分为D7组和D14组,分别在各自的时间点灌注取材L_(4~5)背根神经节,用免疫组织化学方法观察P2X_3受体表达的变化。结果CCI组术侧背根神经节P2X_3阳性神经元数目明显增加,与S组相比,差异有统计学意义(P<0.05);非术侧与S组相比则无统计学差异(P>0.05)。结论坐骨神经损伤后背根神经节P2X_3受体的激活与神经性疼痛的病理过程密切相关。Objective To investigate the changes on expression of P2X3 receptor of rats with chronic neuropathic pain of sciatic nerve in dorsal root ganglion, and to approach the mechanism of chronic neu- rodynia. Methods 24 male SD rats were randomly divided into sham-operated group (S group), sciatic nerve-deligated group( CCI group), and each was devided into D7 subgroup and D14 subgroup, to infuse and take L4_5 dosal root ganglion, and observe the changes on expression of P2X3 receptor with immunohistoehemistry. Results The number of P2X3 receptors immune response-positive neurons in dorsal root ganglion among CCI group had obviously increased. The difference of surgical side was significant( P 〈0.05 ) com- pared to S group;while the difference of non-surgical side was not significant( P〉0.05 ). Condusion The activation of P2X3 receptors in dorsal root ganglion after sciatic nerve injury was correlated closely to pathologic course of neuropathic pain.
