禽传染性支气管炎病毒分离毒株的血清型鉴定与免疫防制研究  被引量:5

摘  要:应用基于单克隆抗体的间接免疫荧光实验(Mab-IFA)和病毒血清中和实验,分别对11个禽传染性支气管炎病毒(IBV)分离毒株进行血清型鉴定。其中3株为Mas型,2株为Ark型,2株为Ark/Mass混合型,其余4株为不可分类型或未知型。将4株不可分类型毒株混合制成IBV分离毒株油乳灭能苗。经SPF鸡免疫攻毒实验证明,这种不可分类型毒株灭能苗与H120弱毒苗合用能够有效抵抗各不可分类型毒株的攻毒感染,保护率为84%~100%。另外,SPF实验鸡经H120和Ark二联苗免疫后对Ark型分离毒株和Ark/Mas混合型分离毒株的保护率为75%~85%。但是,虽经H120弱毒苗免疫SPF鸡则不能抵抗IBV不可分类型毒株。Eleven field isolates of infectious bronchitis virus (IBV)reported in a previous study were serotyped by using a combination of a IBV monoclonal antibodies (Mabs) based indirect fluorescent antibody (IFA) test and by virus neutralization (VN) test in chicken trachealring organ cultures (TROC) with antisera to IBV reference strains.Of 11 isolates serotyped,3 were Massachusetts (Mass),2 were Arkansas (Ark),2 were a combination of Mass and 4 were untypeable.This is the first report of the presence of Ark and/or Arkrelated serotypes and other untypeable IBV isolates in China.An inactivatcd oil emulsion vaccinc made from a pool of 4 untypeable IBV isolates was developed in our laboratory for experimental and field use.Vaccine challenge studies demonstrated that the inactivated vaccine provided 84~100% protection (based on virus isolation from tracheal swabs at days post challenge) in SPF chickens against challenges with the untypeable isolates.IBV H120 and Ark vaccines gave 75~85% protection against challenges with Ark and Ark/Mass isolates.
