中药对衰老大鼠免疫功能的影响及肝细胞的电镜观察

摘　　要：目的:观察制何首乌、北沙参、紫丹参3味中药增强机体免疫力、保肝、抗衰老的作用。方法:将30只Wister雄性大鼠随机分为3组,每组10只。正常对照组:正常大鼠;衰老模型组:大鼠每天颈背部皮下注射D-半乳糖100g·kg^(-1),连续注射40天,形成糖代谢紊乱所致的亚急性衰老模型;中药治疗组:造模的同时,每天2次给予制何首乌、北沙参、紫丹参制成的口服液灌胃。观察大鼠IL-2水平、肝细胞超微结构:结果:衰老模型组大鼠血液中IL-2水平明显降低,肝细胞体积缩小,细胞核和细胞器损伤严重;中药治疗组大鼠血液中IL-2水平升高,肝细胞体积正常,细胞核和细胞器保存良好,接近正常对照组肝细胞的形态。结论:制何首乌、北沙参、紫丹参联合应用可升高机体中IL-2的水平,增强机体免疫力,并且能够改善肝细胞超微结构,抑制肝细胞的凋亡,从而达到抗衰老的目的。Objective:To observe the effect of herbal medicines of Radix Polygoni Mul- tiflori(RPM),Radix Glehniae(RG)and Radix Salviae Miltiorrhizae(RSM)on enhancing immunity,protecting hepatic function and counteracting aging.Methods:Rats were ran- domized into 3 groups,10 in each group.Rat model of sub-acute aging was established with subcutaneous injection of D-galactose 10Og·kg^(-1) for 40 days.The rats in treatment group received oral liquid of RPM,RG and RSM by gastric garage during the modeling,twice per day.Results:Blood interleukin 2(IL-2)level decreased, hepatocytes shrank,and nucleus and organelles were damaged in the model group.In the treatment group,IL-2 increased,hepatocytes became normal,and nucleus and organenes were kept well which was similar to the hepatocytes in the normal group. Conclusion:The combination of RPM,RG and RSM has the anti-aging action by in- creasing the IL-2 level and immunity,improving the ultrastructure of hepatocytes, and inhibiting the apoptosis of hepatoeytes.
