摘 要:目的探讨CD4+CD25+foxp3+调节性T细胞与小鼠GVHD发生的相关性。方法取8~10周龄的SPF级CB6F1小鼠(H-2b/d,♀)30只,随机分为正常对照组、同基因移植组和GVHD组,每组10只,正常对照组不接受照射和移植,同基因移植组小鼠照射后接受CB6F1小鼠骨髓细胞(1×107)和脾细胞(3×107),GVHD组小鼠照射后接受C57BL/6(H-2b,♂)小鼠骨髓细胞(1×107)和脾细胞(3×107)。在发生GVHD的时间点检测3组小鼠脾细胞CD4+CD25+T细胞和foxp3 mRNA的表达。结果正常对照组、同基因移植组和GVHD组小鼠脾细胞中CD4+CD25+T细胞的比例分别为(8.47±1.03)%、(15.40±0.80)%和(24.03±0.85)%,均有显著性差异(P<0.01)。GVHD组小鼠脾细胞foxp3 mRNA的表达水平明显低于正常对照组和同基因移植组,有显著性差异(P<0.001)。结论CD4+CD25+foxp3+调节性T细胞与小鼠GVHD发生呈负相关,发生GVHD时小鼠脾细胞中CD4+CD25+foxp3+调节性T细胞明显减低。Objective To probe the correlation between mice CD4^+CD25^+Foxp3^+ T cells and GVHD of mice allogeneic haemopoietic stem cell transplantation.Methods CB6F1 mice(H-2b/d,♀)were randomized to 3 groups between 8~10 weeks of age,weight was(20± 2)g,control group mice were not given X-ray and transplantation,autologous haemopoietic stem cell transplantation group mice received CB6F1 mice bone marrow cells(1× 107) and spleen cells(3×10^7)after X-ray irradiation,GVHD group mice were given C57BL/6(H-2b,♂)mice bone marrow cells(1 × 107) and spleen cells(3 × 107)after X-ray irradiation.At the time of GVHD happened,3 groups mice spleen cells were prepared,CD4^+CD25^+ T cells were detected with FCM(CD4-pcy5,CD25-FITC),the relative levels of foxp3 mRNA were determined by real-time quantitative RT-PCR in total splenocytes.Results The CD4^+CD25^+T cell of mice splenocytes in control group,auto-HSCT group and GVHD group was(8.47 ± 1.03)%,(15.40 ± 0.80) % and(24.03±0.85)%,respectively(P〈0.01),real-time quantitative RT-PCR showed that the levels of foxp3 mRNA of GVHD group mice splenocytes was lower than control group and auto-HSCT group(P〈0.001).Conclusion There was a negative correlation between CD4^+CD25^+Foxp3^+T cells and GVHD,CD4^+CD25^+foxp3+T cells were lower when GVHD happened.
