Effect of Bone Marrow Mesenchymal Stem Cells on Spleen T Lymphocyte Functions in apoE^(-/-) Mice

摘  要:目的探讨骨髓间充质干细胞对动脉粥样硬化小鼠脾T淋巴细胞功能的影响以探寻新的潜在的治疗动脉粥样硬化的方法和途径。方法分离大鼠骨髓间充质干细胞及野生型和apoE-/-小鼠的脾细胞,单独或混合培养72h,采用流式细胞术检测脾细胞CD4+/CD8+的比值变化;以5mg/mL刀豆蛋白刺激72h,采用非放射性MTS法分析细胞增殖率,用ELISA试剂盒测IFNγ和IL-10的分泌。结果高脂喂养的apoE-/-小鼠脾脏T淋巴细胞的CD4+/CD8+比值、T淋巴细胞增殖率、IFNα的分泌量均显著大于正常饮食的野生型小鼠。经5Gy钴60照射的骨髓间充质干细胞显著地抑制了刀豆蛋白刺激的CD4+/CD8+的比值、T淋巴细胞增殖率、IFNγ的分泌,增加IL-10的分泌,并且对apoE-/-小鼠脾脏中T淋巴细胞的作用显著强于野生型小鼠。结论骨髓间充质干细胞可能通过调节T淋巴细胞的功能而干扰动脉粥样硬化的发生发展。Objective To study the effect of bone marrow mesenchymal stem cells(MSC) on spleen T lym-phocyte functions in apoE^-/-mice.Method Rat bone marrow mesenchymal stem cells and mice splenocytes were isolated and cultured alone or in combination.Flow cytometry was used to assess the ratio of CD4+/CD8+.T lymphocyte proliferation rate was determined by non-radioactivity MTS assay,and IFNγ and IL-10 secretion by ELISA.Results Compared with wide-type C57BL/6J mice,a significant inhibition of CD4+/CD8+ ratio,T cell proliferation rate and pro-inflammatory Th1 cytokine IFNγ secretion was observed in spleen T lymphocytes of apolipoprotein E-knockout(apoE^-/-) mice fed on a high cholesterol diet.MSC irradiated by 5Gy Co60,significantly attenuated concanavalin A(Con A) enhanced CD4+/CD8+ ratio,T cell infiltration and IFNγ secretion in spleen T lymphocytes.The reduction were more significant when cocultured with splenocytes from apoE^-/-mice than from wide-type mice.Whereas an opposite trend was observed with regard to anti-inflammatory Th2 cytokine IL-10 level.Conclusion MSC may exert antiatherosclerotic effects through modulation of T lymphocyte functions.
