摘  要:目的:探讨重组人B7-H1IgV融合蛋白对小鼠骨髓瘤的预防和治疗效果及其对荷瘤小鼠外周血中CD4+CD25+T细胞水平的影响.方法:用重组人B7-H1IgV融合蛋白免疫小鼠,待抗体产生后接种SP2/0肿瘤细胞作为预防组;用重组人B7-H1IgV融合蛋白免疫小鼠,同时接种SP2/0肿瘤细胞作为治疗组.监测小鼠体质量、肿瘤体积的变化及生存期;检测小鼠体内CD4+CD25+T细胞水平.结果:重组人B7-H1IgV融合蛋白在预防组和治疗组中均显示出了较好的抑瘤作用,并可显著降低治疗组小鼠体内CD4+CD25+T细胞水平.结论:重组人B7-H1IgV融合蛋白能够引起小鼠的体液免疫,并抑制SP2/0肿瘤细胞的生长,从而延长荷瘤小鼠的生存期.rhB7-H1IgV融合蛋白肿瘤疫苗有望成为一种肿瘤免疫治疗的候选者.AIM: To analyze the preventive and therapeutic effects of recombinant human B7-HIIgV fusion protein on SP2/0 tumor-bearing mice and observe the change of CD4^+ CD25^+ T cell number in mice. METHODS : The mice of prevention group were inoculated subcutaneously with SP2/0 tumor cells after immunized with rhB7-HllgV protein. The mice of treatment group were inoculated with SP2/0 tumor cells and immunized with rhB7-HllgV protein at the same time. Then we observed the body weight, tumor growth, survival time and the level of CD4^+ CD25^+ T cells in mice. RESULTS : rhB7-H11gV protein delayed tumor growth, prolonged the survival time and decreased the level of CD4^+ CD25^+ T cells in SP2/0 tumor-bearing mice, CONCLUSION: B7-HIIgV fusion protein can activate humoral immune response in mice. It can not only inhibit the tumor growth, but also prolong the survival time of SP2/0 tumor-bearing mice. The rhB7-HllgV fusion protein might be used to develop a new autovaccine in tumor immunotherapy.
