摘 要:目的研究捷安肽素的抗真菌作用机理。方法采用形态学方法和同位素标记法。显微形态观察经捷安肽素处理后的供试真菌的形态学变化。进一步采用14C同位素标记的特异底物"尿苷二磷酸-(14C)-葡萄糖"示踪,研究捷安肽素对真菌(1,3)-β-D-葡聚糖合成酶活性反应的影响。结果显微形态观察表明,经捷安肽素处理后,供试真菌呈现球形膨胀,而且随着捷安肽素作用强度增强,球形膨胀的程度和发生频率也增加,提示捷安肽素的作用位点在真菌细胞壁。同位素标记实验表明,捷安肽素可抑制(1,3)-β-D-葡聚糖合成酶的活性,降低真菌细胞壁合成过程中(1,3)-β-D-葡聚糖的放射性掺入,对疫霉菌和辣椒炭疽病菌(1,3)-β-D-葡聚糖合成酶的抑制率分别达到77.7%和77.5%。结论捷安肽素的抗真菌作用机理为:抑制细胞(1,3)-β-D-葡聚糖合成酶的活性,破坏真菌细胞壁的合成,造成真菌细胞壁组分(1,3)-β-D-葡聚糖的缺损和病原真菌生长抑制。Objective To investigate the antifungal mechanism of Jiean - peptide. Methods The mode of action of Jiean - peptide against 4 phytopathogenic filamentous fungi associated with phytopathy: Phytopluhora sp., Colletotrichum gossypii, Southw Botrytis cinerea and Fusariura oxysporum f. sp. vasinfectura was investigated by the method of morphology and isotope labelling. The (1, 3) - β - D - glucan synthase assay was processed using isotope labelled substrate-urdine diphosphate (^14C) glucose to study the effect of Jiean - peptide on fungal ( 1, 3) - β - D - glucan synthase. Results Micmgraphic observation revealed that the Jiean - peptide made the hyphal expanded and globular. The ( 1, 3) - β- D - glucan synthase assay showed Jiean - peptide could inhibit the activity of ( 1, 3) - β - D - glucan synthase and reduce the radioactivity incorporation of ( 1, 3) - β - D - glucan. The inhibition percentages of Jiean- peptide against Phytopluhora sp. and Colletotrichum gossypii Southw were 77.7 % and 77.5% respectively. Conclusion The antifungal mechanism of Jiean - peptide was the inhibition of enzyme activity of ( 1, 3) - β- D- glucan synthase.
