摘　　要：目的检测系统性红斑狼疮(SLE)患者外周血CD4^+CD25^+FOXP3^+调节性T细胞(Treg)的百分比,并分析其与疾病活动的相关性。方法采用流式细胞仪检测28例SLE患者(其中活动组18例)及22名正常对照组的外周血CD4^+CD25^+FOXP3^+Treg的百分比,同时评估SLE疾病活动指数(SLEDAI)及检测血抗dsDNA水平,分析相关性。结果SLE患者外周血CD4^+CD25^+FOXP3^+Treg占CD4^+ T细胞的百分比较对照组降低(P<0.05),以活动组尤为明显,CD4^+CD25^+FOXP3^+Treg的百分比与SLEDAI呈明显的负相关(P<0.01),同时显示血抗dsDNA阳性组较阴性组患者外周血CD4^+CD25^+FOXP3^+Treg的百分比显著下降(P<0.01)。结论SLE患者外周血CD4^+CD25^+FOXP3^+Treg的百分比在活动期下降为明显,其在SLE的发病机制中可能起到重要的作用。Objective To investigate the frequency of CD4^＋CD25^＋FOXP3^＋Treg cells in the peripheral blood of systemic lupus erythematosus （SLE） patients and its association with disease activity. Methods Peripheral blood mononuclear cells （PBMC） from 28 patients （including 18 active SLE） and 22 healthy controls were counted and stained for CD4, CD25 and intracellular FOXP3. Cells were examined by 3-color staining on the Epics XL-MC and data were analyzed using EXPO32 software. Disease activity was assessed by systemic lupus erythematousus activity index （SLEDAI）. Results The frequency of CD4^＋CD25^＋FOXP3^＋Treg cells was significantly decreased in patients with active SLE compared with patients with inactive SLE and controls [ （1.08±0.43）%, （ 1.58±0.45）% and （1.66±0.34）%, P〈0.01, respectively]. It was inversely correlated with SLEDAI scores （P〈0.01） and serum anti-double-stranded DNA level （P〈0.01）. Conclusion This study demonstrates that CD4^＋CD25^＋FOXP3^＋Treg cells are decreased in the peripheral blood of active SLE patients. It appears that CD4^＋CD25^＋FOXP3^＋Treg cells are powerful modulators of immune system and appears to be important in the pathogenesis of SLE.
