摘  要:目的分析高表达天然抗角蛋白自身抗体的情况下,转基因小鼠肾脏的自身免疫状态。方法选择常规条件下饲养的抗角蛋白自身抗体转基因小鼠为研究对象,Bradford法检测尿蛋白并进行聚乙烯酰氨凝胶(SDS-PAGE)电泳;对肾脏组织进行苏木素-伊红(HE)、过碘酸-雪夫(PAS)染色,并行免疫荧光、电镜等分析。结果与同窝对照小鼠相比,抗角蛋白自身抗体转基因小鼠尿蛋白明显升高(P<0.05),以混合性蛋白尿为主:电镜以系膜增殖性肾小球肾炎为主,荧光显微镜下在系膜区可见IgM类物质呈颗粒状沉积(++~+++)。结论抗角蛋白自身抗体转基因小鼠有发生自身免疫性疾病的倾向,自身抗体在肾脏的沉积可能是疾病发生的原因。Objective To analyze the autoimmune status of kidney in transgenic mice with high serum titer of anti-keratin autoantibody (AK auto Ab). Methods AK auto-Ab transgenic mice were set-up with conventional procedures, and the mice older than 20 weeks were selected for study. Urinary protein was quantified by Bradford method,and SDS-PAGE electrophoresis was used to analyze the type of proteinuria. Specimens of kidney were subjected to HE and PAS stain and immunofluorescence analysis and electron microscopy observation were also performed. Results Compared with littermates, transgenic mice showed higher urinary proteins level (P〈0.05), mainly with mixed proteinuria. Electron microscopy revealed mesangial proliferative glomerulonephritis, and IgM was found to deposit in the glomeruli (++-+++). Conclusion AK auto Ab transgenic mice are prone to develop autoimmune disease and autoantibody deposition in kidney may be an important reason for the pathogenesis.
