摘 要:目的探讨不同剂量芬太尼对乳化异氟醚的催眠半数有效量(ED50)的影响以及芬太尼同乳化异氟醚在大鼠催眠效应的相互作用。方法雄性健康成年SD大鼠125只分为5组,以翻正反射作为终点指标,通过尾静脉穿刺给药,采用序贯法测定芬太尼的ED50,并根据实验结果,用序贯法分别测定合用芬太尼7.2μg/kg(1/4ED50)、14.3μg/kg(1/2ED50)、21.5μg/kg(3/4ED50)以及不用芬太尼时乳化异氟醚的ED50。用Dixon-Mood法计算药物的ED50,用等辐射方法和algebraic分析法分析药物的相互作用。结果各组间大鼠体质量差异无统计学意义;芬太尼的ED50为(28.7±2.1)μg/kg,合用芬太尼7.2μg/kg、14.3μg/kg、21.5μg/kg和不用芬太尼时乳化异氟醚的ED50分别为(0.525±0.032)mL/kg、(0.108±0.019)mL/kg、(0.075±0.011)mL/kg和(0.670±0.054)mL/kg;随着芬太尼剂量增加,乳化异氟醚ED50逐渐呈非线性下降;等辐射和algebraic分析显示芬太尼-乳化异氟醚能产生协同效应。结论芬太尼能增强乳化异氟醚的作用;乳化异氟醚同芬太尼在催眠效应上产生协同效应。Objectives To explore the effect of fentanyl on 50% effective dose (ED50) of emulsified isoflurane for hypnosis, and to define the type of interaction between fentanyl and emulsified isoflurane during rat hypnosis. Methods 125 male adult Sprague-Dawley rats (240-300 g) were divided into 5 groups depending on the drugs administered via the tail vein. The up-an+down sequential allocation technique was used to determine ED50 of fentanyl in the loss of righting reflex. According to above experiment results, the up-an+down sequential allocation technique was also used to determine ED50 of emulsified isoflurane combined with different fentanyl dose of 7.2μg/kg (1/4 ED50), 14. 3 μg/kg (1/2 ED50) or 21.5 μg/kg (3/4 ED50), respectively, and also without the fentanyl in the loss of righting reflex. Fentanyl was administered intravenously for 20 s followed by emulsified isof|urane for 10 s. The interval between the two was 1. 5 min in groups administered with both. ED50 was calculated by using Dixon-Mood method. The isobolographic and algebraic analyses were used to define the type of interaction between fentanyl and emulsified isoflurane. Results The body weight and sex of rats was not significantly different between the 5 groups. ED50 was (28.7±2. 1) μg/kg (26.5-30.8 μg/kg) in the fentanyl group, (0. 525±0. 032) mL/kg in the fentanyl 7. 2 μg/kg+ isoflurane group, (0. 108±0. 019) mL/kg in the fentanyl 14.3 μg/kg+isoflurane group, (0. 075±0. 011) mL/kg in the fentanyl 21.5 μg/kg+isoflurane group and (0. 670±0. 054) mL/kg in the isoflurane group. The isobolographic analysis indicated that ED50 of isoflurane decreased progressively in a non-linear fashion in the presence of increasing dose of fentanyl. The isobolographic and algebraic analyses demonstrated that the combination use of fentanyl and emulsified isoflurane could produce a synergistic effect on hypnosis.
