摘 要:目的:制备表梧儿茶素桔酸酯固体脂质纳米粒并测定其含量。方法:用正交试验法优选处方的组成;用HPLC法测定EGCG的含量,选用LichrospherC18柱(4.6mm×250mm),以甲醇-水(30:70)为流动相,检测波长278nm,流速1.0mL·min^-1。结果:最佳处方为大豆卵磷脂0.2094g,胆固醇0.5578g,表梧儿茶素掊酸酯0.0767g,纯净水30mL;所制得表梧儿茶素梧酸酯固体脂质纳米粒粒径在60-200nm之间,平均粒径120nm,包封率达79.8%,载药量为7.0%。结论:本方工艺可行;建立的高效液相色谱法简便、准确。Objective:To prepare epigallocatechin gallate(EGCG) solid lipid nanoparticles(SLN) and determine the EGGC content in SLN.Method: Orthogonal experiment design was used to optimize the prescription of SLN.The HPLC method was used with a Lichrospher C18(4.6 mm×250 mm) column and a mobile phase consists of methanol-water(30∶70) at the detection wavelength of 278 nm and the flow rate of 1.0 mL·min-1.Result: The best prescription of SLN are: lecithin 0.209 4 g,cholesterol 0.557 8 g,EGCG 0.076 7g and pure water 30 mL.The average diameter of prepared SLN is 120 nm.The entrapment efficiency is 79.8% and ratio of loading drug is 7.0%.Conclusion: The prescription of EGCG SLN is feasible and the established HPLC method is simple,accurate and reliable.
