摘 要:目的:探讨肝癥口服液含药血清对人肝癌BEL-7402细胞系增殖、凋亡、侵袭和转移等生物学行为的影响及其机制。方法:培养技术培养BEL-7402细胞,以原发性肝癌患者服用肝癥口服液前后血清处理肝癌细胞,MTT法测定细胞抑制率,流式细胞术测定细胞凋亡率,通过Boyden小室模型测定其侵袭力、黏附力,细胞迁移实验测定细胞运动能力,同时观察细胞形态,流式细胞术测定肝癌细胞MMP2、TIMP2、integrinβ1、E-cd表达的改变。结果:肝癥口服液含药血清能明显抑制肝癌细胞增殖,促进细胞凋亡,同时抑制肝癌细胞侵袭力、黏附力及运动能力。形态学观察发现,服药后血清组细胞形态较圆,伪足数目较少,MMP2阳性表达细胞减少,TIMP2阳性表达细胞增多,MMP2/TIMP2比值下降;integrinβ1表达增加,E-cd的表达则减低。结论:肝癥口服液含药血清能抑制肝癌BEL-7402细胞增殖,促进细胞凋亡,同时抑制细胞侵袭与转移,其机制可能与直接抑制细胞迁移运动,抑制细胞基质溶解相关基因蛋白MMP2表达,促进TIMP2表达;抑制黏附分子E-cd的表达,促进integrinβ1表达有关。Objective. To study the effect of serum containing Ganzheng solution on biological characters of human hepatoceUular line BEL - 7402 such as proliferation , apoptosis, invasion and metastasis . Methods: Human hepatocellular line BEL - 7402 were cultured by serum of HCC patients before and after taking Ganzheng solution. The effect of serum containing Ganzheng solution on the proliferation of BEL - 7402 ceils was evaluated by MTT, apoptotic rate were observed by FCM, invasion and adhesion assay was conducted in Boyden chamber, cell migration and morphology was also observed. Expression of MMP2 , TIMP2 ,integrinβ1 , and E -cd proteins of human hepatoceUular line BEL -7402 was determined by flow cytometry. Results: Serum containing Ganzheng solution inhibited invasion , adhesion and migration of hepatoceUular BEL - 7402. Meanwhile, expression of MMP2 and E - cd was decreased, the expression of TIMP2 and integrinl31 was increased significantly, MMP 2/TIMP2 ratio was significantly decreased. Morphologically, the cells treated with serum uncontaining Ganzheng solution presented a feature with variously shaped pseudopods, while the ones treated with serum containing Ganzheng solution were kept round with relatively fewer pseudopods. Conclusion: Serum containing Ganzheng solution can inhibite the invasion and adhesion of human hepatoceUular line , which may be the results of decease in MMP 2/TIMP2 ratio and expression of integrinβ1 ,the decrease in expression of E - cd and cell motility.
