HOE642对热缺血死亡大鼠尸体心脏移植的心肌保护作用研究

摘  要:目的在建立热缺血死亡大鼠颈部异位心脏移植模型的基础上,通过对热缺血死亡后不同时段的大鼠心脏添加HOE642(浓度为20μmol/L)灌注,检测供体心肌ATP、SOD和MDA,以观察HOE642对热缺血死亡大鼠尸体心脏心肌的保护作用。方法取清洁级SD雄性大鼠共112只,随机分成7组:每组8只,具体如下:C、正常心脏对照组;S10、S30、S45心脏停搏后10、30及45min取心组;SH10、SH30、SH45心脏停搏后10、30及45min取心并用HOE642灌注组;对实验组大鼠均采用热缺血死亡处理,取得供体后S10、S30、S45组用STH-1液灌注30min,SH10、SH30、SH45组用STH-1液+HOE642(浓度为20μmol/L)灌注30min,再用颈部Cuff法进行异位心脏移植。S10、SH10、S30、SH30于移植后48h取供心标本,S45、SH45组于移植术后取供心标本。然后对标本进行ATP、SOD、MDA的检测。所有数据用SPSS13.0统计软件自动分析。结果大鼠腹主动脉横断放血后9~11(10.11±0.59)min死亡;S10、S30组的ATP含量比SH10、SH30组低,差异有显著性;S45组与SH45组的ATP含量差异无显著性;实验组的SOD含量均比C组低,而MDA含量均比C组高;其中S10、S30组的SOD含量比SH10、SH30组低,而MDA含量则高,差异有显著性。结论热缺血死亡大鼠颈部异位心脏移植模型是进行NHBD的心肌保护研究的理想模型;HOE642能促进热缺血死亡后(30min内)大鼠心脏能量物质ATP合成增加,改善心脏的能量代谢,提高清除氧自由基的能力,同时能减轻脂质过氧化损伤。[Objectives] To set up a heterotopic heart transplantation model of heart transplantation of warm ischemia death rat by infusing HOE642 (concentration 20 μmol/L)into the rat hearts at different stage after the rat death and detecting the ATP, SOD, MDA, so as to study the HOE642 on protecting dead rat myocardium suffered from warm ischemia. [Methods] 112 clearing SD masculinity rats were divided into 7 groups randomly, each group contained 8 rats. The groups were: C, the control group ( normal hearts), S10, S30, S45 (the group of transplanted hearts after 10, 30, 45 min of asystole), SH10, SH30, SH45 (group of transplanted hearts after 10, 30, 45 rain of asystole and infused with HOE642). The rats divided into experimental Sroups were killed totally by warm ischemia, then the donators of the groups(S10,S30,S45) were infused with STH-1 for 30 min, and the dead rats in group SH10,SH30 and SH4 were infused with STH-land HOE642(20 μmol/L)for 30 min, then, heterotopic heart transplantation were processed by the method of neck Cuff. The heart samples of groups (S10, SH10, S30 SH30) were taken after 48 hours of transplantation, red for and the heart samples of groups (S45,SH45) were taken just after transplantation. Then the samples were measu the change of myocardial cell ATP, SOD, MDA. Data was expressed with the software of SPSS13.0. [Result] The rats were discerned death when cardiac electric wave vanished after 9-11 minutes of bloodletting by transseetion of abdominal aorta; Compared with S10, S30 group, the contents of ATP in group SH10, SH30 were higher, the discrepaney was significant, however, there was no significant difference between group S45 and SH45; The contents of SOD in the experimental group were lower then the control group, however, the contents of MDA were higher then the latter, among the total, the SOD in group S10 S30 was lower then which in group SH30, SH30, and the MDA was higher, the difference was significant.
