摘 要:目的研究赤子爱胜蚓组织中脱氧核糖核酸酶1(EWD1)作用于底物的机制。方法采用ZORBAX-Oligo柱-高效液相色谱法(HPLC)、琼脂糖电泳、紫外分光光度法等分析技术,进行了蚯蚓EWD1对环状、线状DNA、单链DNA的降解中间产物和终产物的观察和分析。结果EWD1对所有形式的DNA底物均有降解作用,对单链DNA的降解产物为较均一的、<18bp的寡核苷酸,并显示EWD1无降解RNA的作用。结论蚯蚓组织中发现的EWD1能够分解多种来源的遗传物质,属于脱氧核糖核酸内切酶。Objective To study the mechanism underlying action of a novel deoxyribonuclease (EWD1) from Eisenia foetida. Methods ZORBAX-Oligo column HPLC, agarose electrophoresis and UV spectrophotometer analysis were used to investigate the intermediate and end products of EWD1 processed linear DNA, cyclic DNA and single strand DNA, and also the action of EWD1 on RNA substrates. Results EWD1 was shown to degrade all studied forms of DNA, yielding fairly size-consistent oligo-nucleotides as end-products (〈18 bp). RNA substrates were not degradable with EWD1. Conclusion The EWD1 found in earthworm tissue was found to degrade genetic substance from various sources, although the underlying mechanism needs further exploration. The results indicated EWD1 to be of endonuclease nature (DNases).
