摘 要:目的研究壮肝逐瘀煎对肝纤维化(HF)大鼠Smad3、Smad4、Smad7表达的影响,探讨其抗HF的作用机制。方法Wistar大鼠采用CCl4复合因素法进行HF造模。对HF大鼠予壮肝逐瘀煎灌胃治疗,并以秋水仙碱及大黄虫丸为对照。6周后获取肝组织,苏木精-伊红染色观察HF的程度;免疫组织化学染色、图像分析方法对Smad3、Smad4、Smad7的组织分布进行半定量分析。结果与病理模型组大鼠比较,壮肝逐瘀煎治疗组肝小叶结构趋于正常,纤维间隔明显变薄,肝组织Smad3、Smad4表达显著减少,Smad7表达显著增加。结论壮肝逐瘀煎能够显著改善HF大鼠肝组织的病理变化,具有明显的抗HF作用,其作用机制可能与壮肝逐瘀调控Smad3、Smad4、Smad7的表达有关。Objective: To investigate the effect of ZhuangGanZhuYuJian(ZGZYJ) on expression of Smad3,Smad4 and Smad7 in experimental liver fibrosis in rats, and to explore itsanti - fibrotic molecular mechanism. Methods: The Wistar rat models of liver fibrosis made by Garbon tetrachloride and combined factors were treated with ZGZYJ, Colchicine and DaHuangZheChongWan. After 6 week, the formation of hepatic fibrosis wase observed by using HE stain; the expression of Smad3, Smad4 and Smad7 were detected by using SP immunohistochemistry. Results: In comparison with the model control group, the structure of liver' s lobules became better, fibrous septum changed thinner and the levels of Smad3 and Smad4 were also significantly decreased after treatment with ZGZYJ, whereas the level of Smad7 were significantly increased. Conclusion: ZGZYJ can reverse liver fibrosis in model perfromed by carbon tetrachloride combined factors and its mechanism may be associated with their effect of Regulation and control the expression Smad3, Smad4 and Smad7.
