摘 要:依赖核酸序列的扩增(Nucleic acid sequence based amplification,NASBA)技术是以cDNA为中介体在等温条件下直接扩增单链RNA特异序列的核酸扩增技术,研究探索利用NASBA技术检测草莓斑驳病毒(Strawberry mottle virus,SMoV)。在对14个SMoV分离物序列分析的基础上,设计和筛选出适于SMoV检测的NASBA引物。NASBA检测SMoV的适宜反应温度为40℃。在9份草莓样品上检测SMoV的结果显示,NASBA检测结果与RT-PCR的检测结果一致,这表明初步建立了利用NASBA技术检测SMoV的技术体系。这是利用NASBA检测SMoV的首次报道。Nucleic acid sequence based amplification (NASBA) is a single-step isothermal RNA-specific amplification process that amplifies mRNA in a dsDNA background. In this study, NASBA was applied to detect Strawberry mottle virus (SMoV) in strawberry plants. After the sequences of 14 SMoV isolates were analyzed, the primers for detection of SMoV by NASBA were designed and screened. The suitable temperature of NASBA reaction for detecting SMoV was 40 ℃. Using 9 strawberry samples as materials, the result of detection of SMoV by NASBA was identical with the detection result by RT- PCR. It means that the primary system of detection of SMoV by NASBA was developed in this study. It is the first report of SMoV detection by NASBA.
