摘 要:目的:初步探讨糖尿病大鼠精氨酸血管升压素(AVP)系统的异常及中药黄芪的治疗作用。方法:采用斑点杂交及半定量RT/PCR方法检测链脲佐菌素所致糖尿病大鼠(4周)下丘脑AVP及肾脏皮质V1α受体mRNA表达,同时观察黄芪的影响。结果:糖尿病大鼠血糖及血浆渗透压明显升高,下丘脑AVPmRNA水平是对照组的166倍,该异常与血浆渗透压的变化无明显相关性。肾脏V1α受体mRNA表达也显著上调。黄芪治疗无明显降糖作用,但可纠正糖尿病大鼠过高的血浆渗透压及下丘脑AVP的过度表达,并可导致肾皮质V1αmRNA显著下降。各组动物血钠、心钠素及肾素水平差异均不显著。结论:糖尿病动物存在AVP系统异常(下丘脑AVP表达增多,肾脏皮质受体上调),渗透性和非渗透性因素均可能参与。OBJECTIVE The definite mechanism underlying the changed renal hemodynamics and sodium water metabolism in diabetes mellitus(DM) still remains uncertain. To define the role of AVP system in these changes and to evaluated the therapeutic effects of astragalus memberanaceus (AM), we examined the AVP mRNA expression in hypothalamus and AVP receptor V1α mRNA expression in renal cortex tissue in STZ diabetic rats, with or without the treatment of AM. METHODOLOGY Streptozotocin(STZ) induced diabetic Spragne Dawley rats were applied and divided into diabetic(DM) group and AM treated(AM DM) group(AM at 2.0 gram/day intra peritoneal). The animals were sacrificed at the forth week of observation and plasma, hypothalamus and renal cortex tissue samples were obtained. Plasma sodium, ANP and renin levels were determined by RIA. AVP mRNA expression in hypothalamus was studied by using dot blot analysis and AVP V1α receptor mRNA expression in renal cortex by using semi quantitative RT/PCR method. RESULTS Plasma glucose and osmolality were obviously elevated in both DM and AM DM groups as compared to those of normal rats. Plasma osmolality was found in the normal ranges in AM DM while plasma glucose level not significantly different from the DM group. No difference was found among DM rats, AM DM rats and normal rats for plasma sodium, ANP and renin levels. AVP mRNA level in hypothalamus was found 1.66 times higher in DM rats as compared to the normal rats, while no significant difference was found between AM treated rats and normal control. No correlation between plasma osmolality and hyothalamic AVP mRNA level were found in any group of the animals. A marked increase of renal cortical V1α receptor mRNA expression was observed in all diabetic rats and AM was proved to be able to alleviate this over expression. CONCLUSION STZ induced diabetic rats do have AVP system's abnormalities that might be mediated by the osmotic and the non osmotic mechanisms. AM is of therapeutic effect by ameliorating the changes
