摘 要:目的探讨不同饲料构成对大鼠骨骼肌葡萄糖转运因子-4的影响。方法利用不同饲料(包括基础饲料、高蛋白、高脂肪、高能1与高能2饲料)喂养雄性Wistar大鼠9周,观察不同饲料构成对大鼠体重、脂体比、血糖及胰岛素的影响,并应用蛋白印迹法测定大鼠骨骼肌葡萄糖转运因子-4含量。结果高能饲料1组与高能饲料2组的体重、脂体比、血糖、胰岛素均显著高于基础组、高蛋白组与高脂肪组(P<0.05),而后3组体重、脂体比、血糖、胰岛素比较差异无统计学意义(P>0.05)。高能饲料1组与高能饲料2组葡萄糖转运因子-4含量减少,能量相同的基础组、高蛋白组与高脂肪组含量相近。结论高能饲料可影响机体的葡萄糖代谢。Objective To explore the effects of different diet compositions on glucose transporter 4 in rat skeletal muscle. Methods Male Wistar rats were fed with different diets ( including basic diet, high protein diet, high fat diet, and high calorie 1st and 2nd diet) for nine weeks. The effects of the different diets on weight, the ratio of fat to body weight, blood glucose and insulin were observed. Western blotting technique was used to measure glucose transporter 4 (GLUT4) content in rat skeletal muscle. Results Body weight, the ratio of fat to body weight, blood glucose, insulin in the high calorie 1st and 2nd diet group were obviously higher than that in the groups of basic diet, high protein diet, and high fat diet ( P 〈 0.05), but there was no significant difference between the last three groups ( P 〉 0.05 ). The content of the GLUT4 was decreased in high calorie 1 and 2 groups, and resembled in basic diet, high protein, high fat groups with equal energy. Conclusion High calorie diets can affect the glucose metabolism of the body.
