摘 要:目的研究溪黄草复方对金黄色葡萄球菌生物膜的影响。方法采用试管二倍稀释法测定药物的最低抑菌浓度(MIC),用Brown平板改良法体外复制金黄色葡萄球菌生物膜模型,并检测溪黄草复方的不同MIC浓度对金黄色葡萄球菌生物膜的影响,扫描电镜确认。采用人绒毛膜单层细胞体外培养检测受试药物的毒性作用。结果溪黄草复方对金黄色葡萄球菌的MIC值为128 mg/ml,当MIC值增加至192和256 mg/ml作用于金黄色葡萄球菌生物膜时,其活菌数明显减少。受试药物无体外活细胞毒性作用。结论溪黄草复方对金黄色葡萄球菌生物膜有清除效应,无细胞毒性作用,具备开发应用的潜力,但还需进行药物配方和临床用药研究。Objective To study the effects of rabdosiae serrae compound preparation on staphylococcus aureus biofilm. Methods Minimal inhibitory coh centration (MIC) were tested by doubling dilution, and simulate S. aureus biofilm in vitro be improved Brown plate method. The effects of R. serrae compound preparation in different MIC on S. aureus biofilm were determined and confirmed by scanning electronic microscope (SEM). Single layer cells of human floss film were used to test the toxic effects of the compoud preparation. Results The MIC of S. serrae compound to S. aureus was 128 mg/ml, and the MIC reached 192 mg/ml or 256 mg/ml, the viable count of S. aureus on S. aureus biorilm reduced clearly. The tested preparation had no toxic effects against single layer cells of human floss film. Conclusion R. serrae compound preparation had eliminating effects on the biofilm formed by S. aureus, no toxic effects against human cells in vitro, and a good value to be exploited. The further research of the prescription and the using in clinic was still required.
