养阴清热方对615小鼠HCa-F肝癌凋亡相关蛋白Bcl-2 Bax表达的影响

摘　　要：目的:探讨复方中药抗肿瘤的机制及作用靶点。方法:造模后,荷瘤鼠随机分成6组,分别给生理盐水(空白组)、养阴清热方—安体优Ⅰ号低(ATU-L)、中(ATU-M)、高(ATU-H)3个剂量组灌胃、及阳性对照α-IFN和紫杉醇(PTX)组治疗,第21天处死动物,制备相应标本,以免疫组化SABC法测定瘤组织凋亡相关蛋白Bcl-2、Bax的表达情况。结果:ATU-H组可下调Bcl-2、上调Bax的表达,与空白组及ATU-L组比较,均有显著差异(均P<0.05);ATU-M对Bcl-2、Bax的表达也一定的类似作用,与各组比较,差异不显著(P>0.05);而ATU-L的作用不明显,与空白组差别不大(P>0.05)。结论:安体优Ⅰ号可部分下调615小鼠HCa-F肝癌的凋亡相关蛋白Bcl-2的表达、并上调Bax的表达。Objective ： To discover the anti - tumor machenism and action target of Traditional Chinese Medicine ATU - I on the basis of observing its inhibition of subcutaneous transplanting liver carcinoma（ HCa - F） of ricombinant inbred strain 615 mice. Methods： All the tested carcinoma model mice are grouped into 6 groups which were administered with N. S. （blank group/negative control）, AUT- I low dose （ATU -L）, AUT- I medium dose （ATU -M）, AUT- I high dose （ ATU - H）, α - IFN （ positive control 1 ）, and Pacilitaxel （ pesitive control 2） respectively. The tested mice were killed after 21 days, the expression of tissue apoptosis related proteins： Bcl -2 and Bax were measured through SABC assay. Results ： The expressions of Bcl - 2 and Bax are decreased and increased respectively in ATU - H group and both showed statistically difference compared with those of the blank group and ATU - L group （ P 〈 0.05 ）. Those of the ATU - M group showed similar effectiveness but did not have statistical difference （ P 〉 0.05 ）. Those of the ATU - L group were not obvious and did not have statistical difference compared with the blank group （ P 〉 O. 05）. Conclusion： ATU - I can partly down - regulate the expression of apoptosis related protein Bcl - 2 and up - regulate that of the Bax of the 615 mice with the HCa - F liver carcinoma.
