摘 要:目的比较南充野生芍药叶型、竹叶型半夏的遗传关系。方法使用22条随机引物,对上述两种叶型半夏基因组DNA进行RAPD扩增,比较它们的差异条带。利用Nei,Li方法计算两种半夏之间的遗传相似系数和遗传距离指数。结果22条引物可对南充野生芍药叶型、竹叶型半夏扩增出清晰条带。芍药叶型半夏基因组DNA扩增出157条带,竹叶型半夏基因组DNA扩增出161条带,芍药叶型半夏基因组DNA扩增的差异带3条,竹叶型半夏基因组DNA扩增的差异带7条。它们的遗传相似系数为0.9686;遗传距离指数为0.0314。结论利用RAPD技术证实了野生芍药叶型、竹叶型半夏的遗传关系存在差异,为进一步利用该技术进行半夏分类鉴定等研究奠定了基础。Objective To analyze the genetic relationships between the peony type Pinellla ternate and the bamboo-leave type PineUia ternate in Nanchong area. Methods 22 random oligonucleotide primers were used in RAPD analysis on the genomic DNA of the two types of Pinellia ternate. The RAPD products were analyzed to find out the discrepant bands. The genetic similarity index and the ge- netic distance index were determined by using Nei and Li's methods. Results One hundred and fifty-seven DNA bands were produced from the peony type PineUia ternate. One hundred and sixty-one DNA bands were produced from the bamboo-leave type PineUia ternate. Three diacrepant bands were observed in the peony type Pinellia ternate. Seven diacrepant bands were observed in the bamboo-leave type type PineUia ternate. The genetic similarity index and the genetic distance index of the two types of Pinellla ternate were 0. 9686 and 0. 0314. Conclusion The genetic diversity of the two types of PineUia tenore from Nanchong area was determined by using RAPD, and this technique showed potential value in the identification of PineUia ternate.
