摘 要:目的探讨建立理想的同种异体肢体移植动物模型的方法及技术。方法Wistar大鼠的后肢为移植物,SD大鼠为受体。切取Wistar大鼠的后肢,用UW灌注液,将移植物与宿主的股骨固定,端端吻合坐骨神经、股神经及股动、静脉各一根。手术后长效青霉素抗炎治疗,显微外科术后常规护理,观察移植后宿主及移植物的变化,分析影响异体肢体移植的影响因素和手术技巧。结果共实施68例异体肢体移植手术,其中建立稳定模型前完成24例,成功9例,成功率为37.5%。模型稳定后,又进行大鼠的异体肢体移植44例,成功40例,成功率为90.9%。结论建立稳定的动物实验模型,有利于提高异体肢体移植的存活率,其中熟悉大鼠肢体的解剖是成功的基础,精细稳定的显微外科微创技术是提高血管吻合成功率、保证移植成功的关键。Objective To explore the methods and techniques of establishing a suitable animal model of limb allografting in adult rats. Methods Wistar rats were chosen as donators, and SD rats as receptors. Firstly the hindlimbs of Wistar rats were dissected and washed with UW organ perfusate at 4 ℃ to remove the remaining blood in the grafts before transplanted to the SD rats. The sciatic nerve, femoral nerve, femoral artery and femoral vein were anastomosed end to end in operation. Benzathine benzylpenicillin was given to all the receptors after the operation. The vital signs of the host and the changes of the grafts were observed. Survival rates of the allografts were compared before and after establishment of stable animal models. Results Sixty-eight rat allografts were transplanted in total. Before the model stabilized, 9 out of 24 succeeded (37.5%). After the model stabilized, 40 out of 44 succeeded (90. 9% ). Conclusions Establishment of a stabilized animal model can improve the success rate of allograft. Skills in dissection of rat hindlimbs and sophisticated microsurgical techniques are the most important in avoiding operative hemorrhage and complications and in ensuring successful anglo-anastomosis.
