摘 要:目的:克服一次性PCR假阳性及非特异性条带干扰。方法:建立套式PCR技术,对105份母-脐配对血进行套式PCR、病毒分离、特异性IgM以及特异性IgA测定。结果:母血HCMVDNA阳性6份,脐血3份,母-脐传播率为3/6。3对被证实为母婴垂直传播HCMV的标本中,2对套式PCR、病毒分离、特异性IgM及IgA均阳性,1对套式PCR病毒分离,特异性IgA阳性,但特异性IgM阴性。结论:套式PCR是一种敏感特异、简便快速、能早期诊断HCMV的检测手段,适用于孕妇及胎儿HCMV感染的监测。Objective:To overcome the interruption of false positive and undesirable nonspecific banding with single PCR.Methods:Nested PCR was developed.One hundred and five pairs of maternal-infant umbilical blood were analysed by nested PCR,virus isolation and specific antibodies.Results:Six samples of maternal blood were positive with human cytomegalovirus (HCMV) DNA,3 matched umbilical blood were positive also.The rate of maternal-fetal transmission was 3/6.Of the 3 matched pair blood samples considered as HCMV intrauterine infection,2 pairs were HCMV positive with nested PCR,virus isolation,specific IgM and IgA.One pair was nested PCR,virus isolation,specific IgA positive and specific IgM negative.Conclusions:Nested PCR,being sensitive,specific rapid and simple,was available for the detection of HCM in pregnant women and fetuses.
