摘 要:目的观察酪蛋白(CA)对高碘小鼠甲状腺细胞凋亡的影响。方法将昆明种小鼠随机分为4组:对照(NI)组、高碘(HI)组、高碘+CA1(HI+CA1)组、高碘+CA2(HI+CA2)组,喂养12个月,ELISA法检测血清白细胞介素1β(IL-1β),RT-PCR法检测甲状腺组织Fas、FasL mRNA表达水平,TUNEL法检测甲状腺细胞凋亡。结果与NI组比较,HI组小鼠血清IL-1β水平和甲状腺组织Fas mRNA表达均明显增高(P<0.05或<0.01),且甲状腺滤泡上皮凋亡细胞数也明显增多(P<0.01),而高碘小鼠随着CA摄入量增高,上述改变程度明显降低。结论高碘可通过提高机体IL-1β水平,诱发凋亡调控基因Fas/FasL系统表达改变,促进大鼠甲状腺细胞凋亡。而CA摄入可以部分拮抗高碘引起的上述改变,对滤泡上皮细胞具有一定的保护作用。Objective To observe the effects of casein (CA) on apoptosis of mice thyroid under excessive iodine. Methods Kunming mice were divided into 4 groups: normal control(NI) group, high iodine(HI) group, high iodine plus CA1 (HI + CA1 ) group and high iodine plus CA2 (HI + CA2) group. After twelve months, we examined the levels of IL-1D in the serum by ELISA, thyroid apoptosis by TUNEL, and Fas, FasL mRNA by RT- PCR. Results Compared with that in NI, the levels of IL-1β in the serum, Fas mRNA exprsssion and cell apoptosis of thyroid increased markedly in HI(P 〈 0.05 or 〈 0.01 ), however they decreased in the high iodine plus CA group. Conclusions Iodine excess may elevate IL-1β in the serum and stimulate Fas/FasL system expression of thyroid follicles epithelial cell, leading to thyroid cell apoptosis, which can be protected by CA protein.
