摘 要:目的研究依曲替酸和窄谱中波紫外线(NB-UVB)对培养的正常人角质形成细胞(KC)增殖和维甲酸受体γ(RARγ)mRNA表达的影响。方法用10-6mol/L依曲替酸和/或100mJ/cm2NB-UVB处理正常人KC后,继续培养12h,用RT-PCR和实时荧光定量RT-PCR法分别检测KC增殖和RARγ mRNA表达的改变。结果10-6mol/L依曲替酸或100mJ/cm2NB-UVB处理后12h,KC的增殖均受抑制,抑制率分别为8.7%和14.4%,二者联合作用时,对KC的增殖抑制作用最强,抑制率为19.7%。正常人KC可表达一定量的RARγ mRNA,10-6mol/L依曲替酸处理后,RARγ mRNA的表达增高至正常对照组的2.3倍,100mJ/cm2NB-UVB照射对RARγ mRNA的表达无明显影响,为正常对照组的0.9倍,二者联合作用时,对RARγ mRNA的表达诱导作用最强,为正常对照组的2.8倍。结论依曲替酸和NB-UVB可协同诱导RARγ mRNA表达,并可能参与介导二者对KC增殖的抑制作用。Objective To investigate the changes in cell proliferation and retinoic acid receptor γ(RARγ) mRNA expression in normal human keratinocytes after acitretin treatment and/or narrow-band ultraviolet-B irradiation. Methods Normal human keratinocytes were exposed to irradiation with 100 mJ/cm^2 NB-UVB and/or subsequent 12-hour incubation with 1×10^-6 mol/L acitretin, and the expression of RARγ/mRNA in the cells was examined using RT-PCR and real-time quantitative RT-PCR. Results A 0.9- and a 2.3-fold increase in RARγ/mRNA expression was induced in the cells by exposure to 100 mJ/cm^2 NB-UVB and 10^-6 mol/L acitretin, respectively, and the expression was synergistically enhanced by 2.8-fold after their combined treatment. Conclusion Upregulated expression of RARγ/ mRNA can be associated with keratinocyte growth inhibition after treatment with acitretin and NB-UVB irradiation.
