摘  要:[目的]观察Angptl3在SD大鼠高脂模型中的组织分布与表达水平的差异,探讨其与脂类代谢的关系。[方法]采用高脂饲料诱导高脂动物模型,采用Realtime PCR方法检测脑、肺、心、胃、肝、肾、脂肪等7种组织中Angptl3、LPL的表达水平,分析Angptl3基因表达水平与脂类代谢的关系。[结果]仅在肝、肾组织中检测到Angptl3,其余各组织未见其表达;Angptl3表达水平与血脂水平存在密切关系,随血脂水平升高,其表达增强;LPL在体内各组织均有表达,且表达水平随血脂水平的升高而增强。[结论]在SD大鼠体内Angptl3主要在肝、肾组织中表达,脑、肺、心、胃、脂肪组织表达降低;Angptl3在大鼠体内的组织表达特异性并不随血脂水平的改变而改变,但Angptl3、LPL表达水平均随血脂升高而增强,推测Angptl3可能参与了LPL的表达调控。[Objective]To observe the tissue distribution and expression level of Angptl3 in hyperlipdemia models of rats and explore its mechanism in regulating lipid metabolism.[Methods]The rats were fed with animal feeds which were rich in lipid to induce hyperlipidemia.The method of Realtime PCR was used to detect the expression level of Angptl3 and LPL in brain、lung、heart、gastric wall、liver、kidney and adipose tissues,and the relationship between gene expression level of Angpt13 and lipid metabolism was analyzed.[Results]Angptl3 was only detected in liver and kidney;The expression level of Angpt13 had a close relationship with the level of plasma lipid,and it was enhanced along with the increase of plasma lipid;LPL was detected in every tissue above and its expression level also increased along with the level of plasma lipid.[Conclusions]Angptl3 is expressed in liver and kidney of SD rats exclusively,which would not change with the level of plasma lipid.While the expression level of Angptl3 and LPL are enhanced along with the increase of plasma lipid,which demonstrates that Angpt13 would be likely to regulate the expression of LPL.
