摘 要:目的:研究SMO保存液对犬肾低温保存期间Na+-K+ATPase的影响。方法:建立犬离体肾脏单纯低温保存模型,对照组用0~4℃HTK保存液和UW保存液对供肾进行灌注和保存,试验组用0~4℃自制SMO保存液对供肾进行灌注和保存,分别于24、48、72h后取皮质标本,进行组织病理学观察及检测Na+-K+ATPase的活性。结果:SMO液组所见组织病理学改变,48h和72h比HTK液组轻,在各时间点与UW液组基本相似。Na+-K+ATPase活性,保存各时点,SMO液组与UW液组无明显差异;保存24h和48h,SMO液组与HTK液组无明显差异;保存72h,SMO液组Na+-K+ATPase活性明显高于HTK液组,存在明显差异。结论:SMO保存液在减轻细胞形态学改变和减缓Na+-K+ATPase活性下降方面显著优于HTK保存液,与UW保存液相当。Objective: To study the effect on Na^+-K^+ ATPase in canine kidneys during cold storage with SMO preservation solution. Methods: The model of simple cold storage of isolated canine kidney was set up. Kidneys were flushed and storaged with HTK and UW preservation solution (0~4℃) in the control groups and with SMO preservation solution (0~4℃) in the experimental group, Kidney tissue was sampled at 24,48,72 hrs during cold storage. Histopathology was observed and the activity of Na^+-K^+ ATPase was assayed. Results: The histopathological changes of SMO group were similar to UW group at each time point and less obvious than HTK group at 48h and 72h . At each time point, there was no significant difference in the activity of Na^+-K^+- ATPase between SMO group and UW group. The activity of Na^+-K^+ ATPase in SMO group was higher than that of HTK group and there was significant difference at 72h. Conclusions:SMO was superior to HTK and similar to UW solution with respect to extenuating cell morphology changes and slowing down the degression of the activity of Na^+-K^+- ATPase.
