国产球囊导管制作大鼠主动脉损伤血管内膜增生模型及特性的研究

摘　　要：目的探讨国产2.0 F球囊导管取代进口球囊导管致大鼠主动脉损伤血管内膜增生模型的特点及可行性。方法采用国产2.0 F球囊导管建立大鼠主动脉内皮剥脱模型,分别采用组织形态学和免疫组织化学方法,观察大鼠血管内皮剥脱后内膜增生情况及血管平滑肌细胞(VSMC)表型标志基因SM-肌动蛋白(SMα-actin)、增殖细胞核抗原(PCNA)及Ⅰ型胶原(collagenI)的表达活性。结果采用国产球囊导管反复3次剥脱大鼠胸腹主动脉,与假手术组作比较,术后7 d模型组血管内膜增生不明显,SMα-actin及PCNA表达也均不明显;术后14 d内膜增生较明显,SMα-actin及PCNA表达均明显增强;术后21 d内膜呈进行性弥漫性增生,SM-αactin表达仍明显增高,但PCNA表达不明显。模型组7、14、21 d collagenⅠ表达与假手术组比较,差异均无显著性意义(P>0.05)。结论国产2.0 F球囊导管致大鼠主动脉损伤后,术后14～21 d出现明显的血管内膜增生引起血管再狭窄,其内膜增生主要是由于血管平滑肌细胞增殖所致。表明国产2.0 F球囊导管损伤血管后可取得与进口2F Fogarty球囊导管同样的效果。Objective To investigate the characteristics and feasibilities of intimal hyperplasia model induced by domestic-made 2.0 balloon catheter in place of imported balloon catheter in rats with aortal lesion. Methods The rat model of aortal endothelial denudation was established with domestic balloon catheter; and the intimal hyperplasia after vessel endothelial denudation and the active expression of SMα-actin, PCNA and Collagen I were analyzed with immunohistochemistry and histomophorlogical methods respectively. Results Compared with sham operated group, the intimal hyperplasia in the group redenuded arterial endothelium by domestic balloon catheter was not obvious and the expressions of SMα-actin and PCNA were also not obvious after 7 days; then, hyperplasia began to be observed obviously, and the expression of both SMα-aetin and PCNA were obvious very much after 14 days; and the diffuse hyperplasia was progressive but the expression of SMα-actin was still not obvious after 21 days.The difference of Collagen Ⅰ expression between model group and sham Operated group was not statistic significance （P〉0.05）. Conclusions The vessel restenosis is caused after de-endothelialization from 14 to 21 smooth muscle Forgarty one. days, in which the hyperplasia cell, which shows that domestic is mainly resulted by balloon catheter can be proliferation of vascular in place of imported 2F Forgarty one.
