摘  要:目的探讨姜黄素抑制小鼠黑素瘤作用的可能机制。方法将黑素瘤细胞株B16F10接种于小鼠右大腿外侧皮下,建立荷黑素瘤小鼠模型,于接种后第7天分别予以50和100 mg/kg姜黄素干预,以0.5 mL无血清RPMI 1640培养基腹腔注射为对照组,观察不同处理组瘤体的变化,采用免疫印迹方法检测肿瘤组织核因子κB(NF-κB)p65表达程度,应用半定量逆转录聚合酶链反应方法检测生存素mRNA的水平。结果姜黄素治疗组小鼠瘤重均较肿瘤对照组降低,尤以50 mg/kg姜黄素组为著,抑瘤率可达55.56%(P<0.01);治疗组肿瘤组织NF-κBp65表达水平均较肿瘤对照组降低,50 mg/kg姜黄素组降低63.19%,100 mg/kg姜黄素组降低55.74%,治疗组与肿瘤对照组差异有统计学意义(P<0.01);治疗组生存素mRNA均较肿瘤对照组降低,50mg/kg组降低51.02%,100mg/kg组降低34.69%,治疗组与肿瘤对照组差异有统计学意义(P<0.01);肿瘤组织NF-κB活性与生存素mRNA表达水平呈正相关(r=0.81,P<0.01)。结论姜黄素可能通过抑制NF-κB活化从而下调其下游生存素基因表达来抑制恶性黑素瘤的生长。Objective To investigate the possible mechan-ism of curcumin inhibition to murine melanoma growth. Methods Melanoma cell line B16F10 was injected subcutaneously into the outer side of mouse fight thigh to establish a melanoma-bearing mouse model. Seven days after the establishing the model, these mice were treated with intraperitoneal injection of curcumin at 50 and 100 mg/kg respectively or RPMI 1640 culture medium as control. Fourteen days later, the mice were killed, tumor weight was calculated; the tumor nuclear factor κB activity and survivin mRNA expression were measured by Western blot and RT-PCR, respectively. Results The tumor weight was significantly lower in the curcumin-treated mice than that in the controls (P 〈 0.01 ), with an inhibition rate of 55.56 % by 50 mg/kg of curcumin. The level of NF-κB activity and survivin mRNA expression were decreased by 63.19% and 51.02%, respectively, by curcumin of 50 mg/kg, 55.74% and 34.69%, respectively, by curcumin of 100 mg/kg (all P 〈 0.01 ), as compared with the culture medium. There was a positive correlation between the tumor NF-κB activity and survivin mRNA expression (r= 0.81, P 〈 0.01 ). Conclusion Curcumin may inhibit the growth of malignant melanoma by suppressing NF-κB activation and down-regulating survivin gene expression.
