摘  要:目的初步探讨应用血清蛋白组学技术建立寻常性银屑病相关蛋白的优化筛选方法,筛选寻常性银屑病的相关蛋白,期望从中发现有意义的寻常性银屑病血清标志物。方法取寻常性银屑病患者及正常对照血清,应用ProteoExtract Albumin/IgG Removal Kit纯化血清蛋白,去除白蛋白和IgG,采用固相pH梯度(IPG)、等电聚焦(IEF)为第一向、SDS-PAGE垂自电泳为第二向的双向凝胶电泳(2-DE)和质谱分析鉴定差异蛋白。结果获得重复性和分辨率较好的血清双向凝胶电泳图谱,鉴定得到3个仅在银屑病组血清中表达和9个与对照组血清表达有差异的蛋白质。结论在寻常性银屑病患者血清与健康人血清中存在差异表达蛋白质,利用蛋白质组学这一高通量的筛查技术可望从中发现与寻常性银屑病相关的标志物。Objective To investigate the establishment of better screening methods of associated proteins in human psoriasis vulgaris by serological proteomics technologies, and to screen the differential express proteins related with psoriasis vulgaris. Meth- ods Samples of peripheral blood were collected from newly diagnosed psoriasis vulgaris patients in the clinic, and 20 sex and age matched healthy persons. Removaling serum albumin and IgG were achieved by filtering with ProteoExtract Albumin/IgG, 2-DE technique,including isoelectric focusing (IEF) with an immobilized pH gradient (IPG) and SDS-PAGE, were used, associated with Mass Spectrometry(MS) to identify the different protein spots. Results Electrophoresis figures with high resolution and reproduc- ibility were obtained. Three differential expression proteins were found only in the serum from psoriasis vulgaris patients, while nine other differential proteins expressing from healthy volunteers. Conchlsion Protein expression is differential in serum from the pso- riasis vulgaris patients and health volunteers. It is hopeful to find the biomarkers related psoriasis vulgaris using proteomics.
