摘  要:目的探讨五种化学消毒剂在灭活病毒试验中选择中和剂的试验。方法采用细胞感染法和噬斑计数法(Pfu)观察了消毒剂灭活病毒试验中所用中和剂的效果。两种病毒灭活试验中均以噬斑数计算中和剂、中和产物、稀释液三组间误差率。结果在流感病毒中和剂鉴定试验中,所有消毒剂试验组所得的误差率均小于15%;说明该方法选择的中和剂可用于该病毒灭活试验中残余物去除。在脊髓灰质炎病毒中和剂鉴定试验中,除邻苯二甲醛之外,另外4种消毒剂所得的三组间平均误差率均大于15%,说明该中和剂选择试验程序不适合所试验的消毒剂灭活脊髓灰质炎病毒试验。结论在消毒剂灭活病毒试验中,培养细胞对中和剂及其产物较试验病毒更为敏感,该中和剂对MDCK影响较对VERO的影响小。本试验方法可用于流感病毒灭活试验的中和剂鉴定试验。Objective To investigate neutralizer selection test in test for inactivating virus by five kinds of chemical disinfectants.Methods Method of virus infection of cells and plaque forming unit(PFU) counting method were used to observe the efficacy of the neutralizer used in test for inactivating the virus by disinfectant.The plaque forming unit count was used to calculate the error rates between neutralizer group,neutralizing product group and diluent group.Results In the neutralizer verification test for influenza virus,the error rates of all disinfectant test groups were less than 15%,indicating that the neutralizer selected could be used for removing the residual disinfectant in this virus inactivation test.In the neutralizer verification test for poliovirus,besides ortho-phthalaldehyde,in the other 4 disinfectants tests,the average error rates between the three groups were 〉15%,indicating that the neutralizer selection test procedures were not suitable for the test for inactivation of poliovirus by the disinfectant tested.Conclusions In virucidal test for disinfectant,the cultured cells are more sensitive to neutralizer and its neutralizing product than the test virus is.The influence of the neutralizer on MDCK cell line is less than on VERO cell line.The tsst method can be used in neutralizer verification test for influenza inactivation test.
