摘  要:目的观察大鼠脑微血管内皮细胞缺血再灌注损伤细胞间黏附分子-1(ICAM-1)表达的影响,探讨清开灵有效组分猪胆酸阻抑脑缺血炎症反应的作用途径。方法体外培养大鼠脑微血管内皮细胞,氧糖剥夺法模拟缺血再灌注损伤,采用免疫细胞化学染色和RT-PCR方法半定量检测ICAM-1蛋白和mRNA的表达。结果与正常组比较,模型组ICAM-1蛋白和mRNA的表达明显增高(P<0.01);与模型组比,猪胆酸显著降低ICAM-1的表达(P<0.05)。结论猪胆酸可能通过干预ICAM-1表达而阻抑脑缺血损伤炎症反应。Objective To observe the influence of hyocholic acid on the expression of intercellular adhesion molecule-1 ( ICAM-1 ) of brain microvascular endothelial cells (MVEC) in rats after ischemia and reperfusion injury and to investigate the mechanism of hyocholic acid for inhibiting the inflammatory reaction in cerebral ischemia. Methods The rat cerebral MVEC were cultured in vitro and the ischemia and reperfusion injury was simulated by using oxygen and glucose deprivation (OGD). The expressions of ICAM-1 and mRNA were detected semiquantitatively by immunocytochemical stain and RT-PCR. Results The expressions of ICAM-1 and mRNA were obviously increased in the model group than those in the normal group (P 〈0.01). Hyocholic acid reduced significantly the expression of ICAM-1 compared with the outcome of the model group (P 〈 0.05 ). Conclusion Hyocholic acid may inhibit the inflammatory reaction in cerebral ischemia through intervening the expression of ICAM-1.
