摘 要:目的:预测HBV基因编码的microRNA分子前体.方法:采用VMir软件,对HBV全基因进行扫描,通过对所得序列的评分及结构分析,得到HBV编码的microRNA的候选前体分子;通过Northern blot实验,对预测结果进行初步验证.结果:通过VMir的预测,得到3个HBV可能编码的microR-NA分子前体,即MD6,MD20,MR31;通过Northern blot实验,只有针对于MD17的探针杂交后出现特异性条带.结论:MD17可能为HBV基因组编码的microRNA前体分子.AIM: To predict HBV genome encoded microRNA precursor.METHODS: HBV complete genome was screened with software, VMir, and the sequences obtained were scored and analyzed in their structure. Finally the sequences, which are potential microRNA precursor encoded by HBV genome, were got. Then the results above were proved by Northern blot assay. RESULTS: With the help of VMir, we got three sequences, MD6, MD17, MR31, which may be potential precursor microRNAs encoded by HBV genome; Northern blot assay indicated that specific lane appeared only when MD17 specific probe was used. CONCLUSION: MD17 may be the microRNA precursor encoded by HBV genome.
