摘  要:目的:观察并检测5-氮胞苷(5-Aza)诱导大鼠脂肪间充质干细胞(AMSCs)分化成肌细胞的过程中,细胞形态及相关基因、蛋白的表达情况,探讨5-Aza对大鼠AMSCs分化的诱导作用.方法:分离大鼠AMSCs,在含有10μmol/L5-Aza的培养液中培养,观察细胞形态的变化,并通过逆转录-聚合酶链式反应(RT-PCR)检测诱导后MyoG的表达,免疫细胞化学染色法检测诱导后Desmin和α-Sarcometric Actin的表达.结果:用10μmol/L5-Aza诱导后,细胞形态逐渐伸展变长,呈现肌细胞形态;RT-PCR结果显示,诱导后15h左右开始表达MyoG;免疫细胞化学染色结果显示,诱导10d后Desmin染色呈弱阳性,14d阳性程度增强,且此时部分细胞开始表达α-sarcomeric Ac-tin,并随时间延长表达增强,至28d呈强阳性.结论:采用10μmol/L5-Aza可以诱导大鼠AMSCs向肌细胞分化.AIM: To explore the effects of 5-Aza on the expressions of MyoG, Desmin and α-Sarcometric Actin of rat adipose tissue-derived mesenchymal stem cells ( AMSCs ). METHODS : Rat AMSCs were cultured in vitro, and then were incubated with 10 μmol/L 5-Aza. The expression of MyoG was evaluated with reverse transcription-polymerase chain reaction (RT-PCR) method, the antigen expressions of Desmin and α-Sarcometric Actin with immunohistochemistry and the cell morphological changes were observed. RESULTS: After induced by 10 μmol/L 5-Aza, some cells were obviously enlarged and began to express MyoG at the 15^th hour. AMSCs began to express Desmin at the 10^th day and reached the higher lenel at the 14^th day. AMSCs began to express et-Sarcomeric Actin at the 14^th day and up to the highest level at the 28^th day. CONCLUSION: Rat AMSCs can be induced to differentiate into myoblasts and gradually express MyoG, Desmin, α-Sarcomeric Actin.
