摘 要:为探讨小麦PPO基因变异对籽粒PPO活性的影响,研究了191份小麦及其近缘种属材料的籽粒PPO活性及TaPPO-D1基因的变异特点。结果表明,PPO活性在所研究的材料中变异大,特别在普通小麦中,PPO活性分布很广(10-288 U·g^-1.min^-1)。但总体来说,PPO活性处于中等偏低的水平。六倍体小麦的PPO活性高于粗山羊草;野生二粒小麦的平均活性最低。在小麦及其近缘种属中,TaPPO-D1基因存在丰富的变异,其中以D类型居多;一些类型与PPO活性关系密切,如A、B、E、F类型与高PPO活性有关,而C、H、I、K多分布在中低活性的材料中,野生二粒小麦则缺失该基因。对D、I两种类型的PCR产物测序表明,D类型和TaPPO-D1b等位基因相同,而I类型则和TaPPO-D1a相同。For characterizing PPO gene and PPO activity,TaPPO-D1 variations and PPO activity were investigated in common wheat and other species related to wheat.The results indicated that there were very large variations in PPO activity,especially in common wheat with a range of activity from 10 to 288 U·g^-1·min^-1.However,middle to low level of PPO activity occurred in most samples.Hexapliod wheat(AABBDD) showed higher PPO activity than Aegilops tauschii(DD) and the lowest average PPO activity was found in wild emmer wheat(T.turgidum var.dicoccoides,AABB).Rich allelic variations were identified in common wheat and its related species.Of these alleles,D-type was prevalent as compared with others.Some TaPPO-D1 alleles had a close relationship with PPO activity in seeds,but it was null in wild emmer wheat with very low activity.Based on the results of sequence,the alleles of D-type and I-type corresponded to TaPPO-D1b and TaPPO-D1a,respectively.
