摘 要:目的检测藻酸盐微球对间充质干细胞生长的影响,探讨其在骨组织工程中可能的应用。方法应用滴注法制备藻酸盐微球,检测不同浓度的藻酸盐微球pH值的动态变化及通过MTT法和流式细胞仪检测藻酸盐微球溶液中间充质干细胞的生长情况,评价藻酸盐微球对细胞的影响。结果间充质干细胞可黏附在藻酸盐微球上生长,浓度为0·02g/L、0·2g/L和2·0g/L的藻酸盐微球溶液的pH值分别在7·20、7·20和6·70左右波动,MTT法检测这3个浓度组同细胞对照组D(490)值几乎无差异,提示对间充质干细胞的生长影响不具有统计学意义。细胞周期分析显示,3d时各组S期细胞比例均等于或大于对照组,且随着藻酸盐微球浓度的增加而增加;7d时各组S期细胞比例与对照组相对差值在±3.15之间,0·2g/L组比例最高,达24·40%。结论浓度等于或低于2·0g/L的藻酸盐微球溶液对间充质干细胞生长影响很小,适合与之共培养进行有关研究。Objective To when coculture and explore their investigate the effect of alginate beads on mesenchymal stem possible application in bone tissue engineering. Methods were produced by dropping. The development of pH value and generational situation of MSCs in cells (MSCs) Alginate beads alginate beads solution at different concentrations was detected respectively by pH meter, MTT and flow cytometry. MSCs attached on alginate beads. The pH values of 0.02, 0.2 and 2.0 g/L alginate beads solution ated around 7.20, 7.20 and 6.70 respectively. The OD value detected by MTT method in above 3 similar to blank cell control group. The analysis of cell cycle showed that the rates of S stage cells in Results were fluctu-groups was alginate solution were equal to or larger than the control group on 3^rd day and these rates on 7^th day in alginate solution were similar to the control group. Conclusion The results suggest that alginat 2.0 g/L has less effects on the growth of MSCs and reaches the microenvironment e beads solution lower than needs of MSCs.
