摘  要:目的:观察长期游泳运动后不同性别大鼠心肌丙二醛含量、谷胱甘肽含量和谷胱甘肽过氧化物酶活性的变化,探讨长期游泳运动对大鼠心肌脂质过氧化应激反应和抗过氧化作用影响的性别差异。方法:实验于2006-03/10在江苏大学医学院实验室完成。①实验分组:选择3月龄Sprague-Dawley大鼠40只,随机分为雄性静息组、雄性运动组、雌性静息组和雌性运动组,每组10只。②实验方法:雄性运动组及雌性运动组在水深50cm和水温(35±1)℃的玻璃游泳池内游泳,每周连续游泳5d(休息2d),1次/d,每次游泳持续时间为:第1周30min,第2周1h,第3周开始固定为2h,共持续3个月。雄性静息组及雌性静息组除不进行运动外,其余处理与雄性运动组及雌性运动组相同。③评估指标:在实验期满后麻醉下处死各组大鼠,取左心室心肌,采用改良的硫代巴比妥法测定丙二醛浓度;采用二硫二硝基甲酸反应法测定谷胱甘肽浓度;采用酶促反应谷胱甘肽消耗法测定谷胱甘肽过氧化物酶活性。结果:纳入动物40只,均进入结果分析。①丙二醛含量:雄性运动组与雄性静息组之间差异无显著性,而雌性运动组显著高于雌性静息组(P<0.01);雌性静息组与雄性静息组之间差异无显著性,但雌性运动组显著高于雄性运动组(P<0.01)。②谷胱甘肽含量:雄性运动组显著高于雄性静息组(P<0.01),雌性运动组与雌性静息组之间差异无显著性;雌性静息组与雄性静息组之间差异无显著性,雄性运动组显著高于雌性运动组(P<0.01)。③谷胱甘肽过氧化物酶活性:雄性运动组显著高于雄性静息组(P<0.01),而雌性运动组与雌性静息组之间差异无显著性;雌性静息组与雄性静息组之间差异无显著性,但雄性运动组显著高于雌性运动组(P<0.01)。结论:长期游泳运动对心肌过氧化应激和抗过氧化能力的影响存在性别差异,雄性心肌的抗氧化能力对长期游泳运�AIM: To observe the changes of myocardial malondialdehyde (MDA) content, glutathione (GSH) content and glutathione peroxidase (GSH-Px) activity after long-term swimming exercise for investigating the gender differences in the effects of long-term swimming exercise on lipid peroxidation stress and anti-oxidation reaction in rat cardiac muscles. METHODS: The experiment was conducted in the laboratory of School of Medicine, Jiangsu University between March and October in 2006. ①Forty Spraque-Dawley rats of 3 months were selected and divided into four groups at random (n = 10): a male sedentary group (MSG), a male exercise group (MEG), a female sedentary group (FSG) and a female exercise group (FEG). ②The rats in the MEG and the FEG swam in a glass swimming pool filled with water to 50 cm depth at (35±1) ℃, once per day, continuous 5 days per week, lasting for 3 months. The daily training lasted for 30 minutes in the 1^st week, 1 hour in the 2^nd week and 2 hours from the 3^rd week. The rats in the MSG and the FSG remained sedentary in their cages throughout the experiment and received approximately the same handling as the exercised rats. ③At the end of the experimental period, all the rats were anesthetized to death, then the MDA content, GSH content, and GSH-Px activity in the rat cardiac muscle were measured with modified thiobarbituric acid chromatometry, disulfide dinitro-formic acid reaction, and GSH consumption enzymatic reaction, respectively. RESULTS: A total of 40 rats were involved in the analysis of results.①No significant differences in the contents of MDA were found between the MEG and the MSG, but the MDA contents in the FEG were significantly higher than those in the FSG (P 〈 0.01).
