摘　　要：目的:研究半乳凝集素-3(Gal3)的表达对乳腺癌细胞MCF-7细胞凋亡的影响。方法:构建Gal3-siRNA的特异表达载体,转染乳腺癌细胞MCF-7,通过建立由siRNA介导的Gal3-knockdown稳定细胞株来研究半乳凝集素-3表达下调对肿瘤细胞凋亡的影响。结果:利用设计的Gal3-siRNA能够使细胞中半乳凝集素-3的表达降低90%左右;当用凋亡诱导剂处理时,Gal3-knockdown稳定细胞株的凋亡率比野生型细胞株高出近20%。结论:在MCF-7乳腺癌细胞中,半乳凝集素-3具有抑制肿瘤细胞凋亡的功能。Objective： To investigate the effect of galectin3（Gal3） expression on apoptosis in MCF-7 cell lines. Methods： a Ga/3-siRNA expression plasmid was constructed and transfected into MCF-7 cell. Through the stabled-transfected, the siRNA-mediated Ga/3-knockdown cell lines were obtained, and the effect in apoptosis was detected. Results： The results showed that the galectin3 expression was knocked down 90%, and the rate of apoptosis was higher 20% than the wild type tumor cell induced by drug. Conclusion： It is demonstrated that Gal3 exibits an anti-apoptotic activity in MCF-7.
