摘  要:目的对新型组织工程神经支架材料——胶原-明胶(CG)支架材料用不同的交联方法进行改性,研究改性后材料的细胞毒性。方法用紫外线照射、戊二醛浸泡、京尼平交联三种交联法对CG支架材料进行交联改性,遵照GB/T16886/ISO10993医疗器械生物学评价之体外细胞毒性实验原则,采用国际标准的两种实验方法,选用建系的L929小鼠成纤维细胞对改性后的支架材料进行体外细胞毒性实验。结果三种交联法改性后的CG支架材料对体外培养的细胞形态不构成损害,对其生长和增殖无明显抑制作用,细胞毒性为0~1级。结论用三种交联法改性后的CG支架材料均无明显细胞毒性。Objective To modify the new reparing material of nerve tissue engineering-collagen- gelatin (CG) stent biomaterial with different cross-linking methods and study the cytotoxicity of this modified biomaterial. Methods Ultraviolet rays, glutaraldehyde,genipin were used for crosslinking the CG biomaterial. According to the experimental principle of GB/T16886- ISO10993 on medical apparatus,and two international standard experimental methods were adopted in the study. The L929 fibroblasts were used in this study to establish cell proliferation. Results Crosslinked CG biomaterial might not be harmful to the morphology of the L929 cell. The growth and proliferation of the L929 cell could not be inhibited significantly. The cytotoxicity score of the biomaterial was in grade 0 - 1. Conclusion It is demonstrated that the crosslinked CG biomaterial shows no obvious cytotoxicity.
