摘 要:以粤香占和丰美占两种水稻为指示植物,通过透射电镜、扫描电镜以及彗星实验,在根尖细胞超微结构和DNA分子水平上研究了重金属元素Cd、苄嘧磺隆(Bensulfuron Methyl,BSM)复合污染对水稻的毒理学机制。电镜分析结果表明BSM和Cd复合污染能够破坏水稻根尖的细胞结构,使表皮细胞破裂,核仁解体。对丰美占水稻进行彗星实验研究,并用CASP软件对图像进行分析。研究结果表明,随着Cd2+浓度的增加,DNA的迁移程度逐渐增加,也就意味着DNA的受损程度逐渐增加。当Cd浓度为10mg·L-1时,0.1mg·L-1的BSM会使OliveTM等参数降低,这说明0.1mg·L-1的BSM能够在一定程度上缓解Cd对水稻叶片DNA的损伤。To investigate the effects of co-contamination of bensulfuron methyl and cadmium on rice at cellular and molecular levels, transmission electron microscope (TEM), scanning electron microscope (SEM) and comet assay were used. Two rice cuhivars, Yue Xiangzhan and Feng Meizhan, were employed as the indicator plants. TEM and SEM were used to assess the toxicity of Cd and BSM to rice at the cellular level. The result indicated that BSM and Cd could destroy cellular structure, disrupt epidermic cells and result in nucleolus resolution. The comet assay was used to investigate the toxicity of Cd and BSM to Feng Meizhan at molecular level by analyzing the photograph with CASP. The result indicated that there was a dose dependent increase in DNA damages with the increasing concentrations of Cd in the solution. When 0. 1 mg·L^-1 BSM was added in the solution with 10 mg·L^-1 Cd, the Olive TM was decreased, indicating the toxicity of Cd to the rice DNA could be relieved when the 0. 1 mg·L^-1 BSM was supplied.
